Synthesis of stereocontrolled α,α-difluoro-β-hydroxycarbonyl materials

Synthesis and synthetic utilities of stereocontrolled α,α-difluoro-β-hydroxy-γ,δ-unsaturated carbonyl compounds via enzymatic resolution with lipase PS (Pseudomonas cepacia, Amano Pharmaceutical Co. Ltd.) or lipase MY (Candida rugosa, Meito Sangyo Co. Ltd.) were described, and then the absolute configuration of obtained chiral materials was determined by the modified Mosher's method.     

Fluorescence modification of Gb3 oligosaccharide and rapid synthesis of oligosaccharide moieties using fluorous protective group

The use of the bisfluorous chain-type propanoyl (Bfp) group as a fluorous protective group made it possible to rapidly synthesize the Gb2 and Gb3 oligosaccharide derivatives by a simple fluorous-organic extraction purification. Furthermore, the fluorescence-labeled Gb2 and Gb3 oligosaccharides were prepared as a potential Vero Toxins detecting reagent.     

Isotachophoretic determination of mobility and pKa by means of computer simulation : V. Evaluation of mo and pKa of twenty-eight dipeptides and assessment of separability

Isotachophoretic qualitative indices, R_E, for twenty-eight dipeptides were measured in the range pH 7.4–9.6. The absolute mobility, m_o, and pK_a values were evaluated by the use of the least-squares method, utilizing a simulation of the isotachophoretic steady state. The m_o values were newly evaluated and the pK_a values were in good agreement with literature values. By comparison of the evaluated m_o and pK_a values of the dipeptides with those of the constituent amino acids, simple relationships were found which may be used to estimate the m_o and pK_a values of other dipeptides. The separability of the dipeptides was also evaluated by considering the differences between their simulated effective mobilities. It is concluded that isotachophoresis is very convenient for the separation of dipeptides and their constituent amino acids.     

Total synthesis of (+)-pederine. A simple synthetic method for N-(1-methoxyalkyl)amides

A mild one-pot method for the synthesis of acyclic N-(1-methoxyalkyl)amides starting from carboxylic acids and methyl imidates had been developed and applied to the total synthesis of the insect poison pederine $\text{1}$.     

Novel Z-selective head-to-head dimerization of terminal alkynes catalyzed by lanthanide half-metallocene complexes

Various terminal alkynes have been cleanly dimerized into the corresponding head-to-head (Z)-enynes by use of the half-metallocene lutetium alkyl complexes Me2Si(C5Me4)(NAr)Lu(CH2SiMe3)(THF) (Ar = Ph, C6H3Me2-2,6, C6H2Me3-2,4,6) as catalysts. Aromatic C-Cl, C-Br, and C-I bonds, which are known to be extremely susceptible to reductive cleavage by transition metals, survived in the present reactions. The corresponding dimeric alkynide species [Me2Si(C5Me4)(NAr)Lu(mu-CCR)]2 are thought to be the true catalysts, some of which have been isolated and structurally characterized. These alkynide species were thermally stable and soluble at the reaction temperatures (80-110 degrees C), but they precipitated upon cooling to room temperature after completion of the reaction. Therefore, this catalyst system works homogeneously but can be separated and reused, thus constituting the first example of a recyclable catalyst system for the dimerization of terminal alkynes and also the first example of (Z)-selective head-to-head dimerization of aromatic terminal alkynes.     

Heteroatom-guided torquoselective olefination of alpha-oxy and alpha-amino ketones via ynolates

Ynolates were found to react with alpha-alkoxy-, alpha-siloxy-, and alpha-aryloxyketones at room temperature to afford tetrasubstituted olefins with high Z selectivity. Since the geometrical selectivity was determined in the ring opening of the beta-lactone enolate intermediates, the torquoselectivity was controlled by the ethereal oxygen atoms. From experimental and theoretical studies, the high Z selectivity is induced by orbital and steric interactions rather than by chelation. In a similar manner, alpha-dialkylamino ketones provided olefins with excellent Z selectivity. These products can be easily converted into multisubstituted butenolides and gamma-butyrolactams in good yield.     

Solvent Effects on the Absorption Spectrum of the Product and the Equilibrium Constant for the Proton Transfer Reaction from 1-Phenacylquinolinium Bromide to Amines

The equilibrium constants, K ₂, have been determined for the proton-transfer reactions of 1-phenacylquinolinium ion, PHQ⁺, with several amines {triethylamine (TEA), N,N,N′,N′-tetramethylethylenediamine (ED), N,N,N′, N′-tetramethylpropanediamine (PD), N,N,N′,N′-tetramethylbutanediamine (BD), and 1,8-bis(dimethylamino-naphthalene (DMAN)} in acetonitrile (AN), AN-tetrahydrofuran (THF) and AN-ethanol (EtOH) mixtures. The reaction was followed spectrophotometrically using a stopped-flow technique. The K ₂ value decreased for DMAN and increased for TEA with increasing vol-% of THF in AN-THF mixtures. The changes in the K ₂ value for ED, PD and BD changed in the order: ED, PD and BD from a pattern similar to TEA to a pattern similar to DMAN. The change in the K ₂ value for DMAN with increasing vol-% of THF in AN-THF mixtures was explained by the effect of polarity on the stability of P⁻Q⁺ (the deprotonated product of PHQ⁺). The effect of THF on the K ₂ value is consistent with that of the peak wavelength of the absorption spectrum of P⁻Q⁺. The change in the K ₂ value for TEA, ED, PD and BD depended on the structures of the protonated bases, one of the products for this reaction. The effect of EtOH on the K ₂ value for DMAN was examined in ternary EtOH-THF-AN mixtures that contain different amounts of EtOH and whose relative permittivities were adjusted to that of EtOH. The K ₂ value increased with increasing vol-% of EtOH because of the stabilization of P⁻Q⁺ upon the formation of the hydrogen-bonded complex with EtOH. The absorption spectrum of P⁻Q⁺ demonstrated a blue shift as the vol-% of EtOH increased.
